It depends on how Kishi chose to draw them, as in what tact he took.

If they were overly obvious objects of fanservice busting out of Sakura's too-small shirt and ending up becoming one of the biggest reasons behind her fandom, then her character probably wouldn't have that same down-to-earth feel that it has for me. This isn't necessarily due to the change in her appearance in and of itself, so much as how the mangaka is setting her up to be viewed.

Also, they would make it so some of my favorite portions of her animated fights/conflicts had to be completely revamped, as it's incredibly unrealistic to think that someone with such large breasts is going to be able to squeeze and dash as lithely through all of Sasori's puppets. When you only have a hairs' breadth of free space, adding in a huge bust is pretty much going to cripple your acrobatics. And, if it didn't, then it would still make her dynamic fighting skills less impressive to watch for me as they would seem less important athletically as compared to being cheap fanservice (considering how she would be moving and how such movement would affect giant breasts). It's not that I can't enjoy fanservice, but certain episodes were such great instances of art and animation in how they utilized the potential of her slender yet strong, flexible form.

When all is said and done, I wouldn't dislike Sakura, if she were to gain huge breasts. But, their presence has the potential to take away some of my favorite aspects of her character which aren't actually related to her appearance, so much as her character's tone and ability. So, Sakura gaining huge breasts could end up making me lose a good deal of my interest in her character.
