The Killer gets his dual pistols and 60 shots. He has the flight boots The Professor gives him, but he's limited to subsonic speeds. Konoha gets everyone that's currently alive and still a part of its forces (meaning no Sasuke, Itachi, Orochimaru, etc.).
Arena: Konoha
Win: The Killer must kill Tsunade. Konoha must kill The Killer. There is no ringout.
Bloodlust: Yes, but nothing out of character meaning Kakashi wouldn't just MS him off the bat, Naruto cannot go KN4 immediately, etc.
Time Limit: 24 hours
Prep-time: Konoha is aware that someone will try and kill Tsunade, but have no info on his abilities or where he's coming from, nor what time. The Killer does not have knowledge of the Konoha ninjas' abilities.
Everyone knows Konoha's abilities, so here's a brief rundown on The Killer:
1. He apparently has the subconscious knowledge of how to kill anything. With regular pistols, he was fighting off hordes of physically-superior superheroes, and winning.
2. He can't miss. Ever. On his very first time picking up a gun (or any weapon, really), he shot the wings off a fly.
3. Going back to his subconscious knowledge bit, he states that unlike other people who have more flashy powers like flight or superstrength, his ability is very simple: he makes people stop breathing. He killed Shithead, who was literally a shit elemental composed of the shit of the 666 most evil people who ever lived. He did this by shoving him face first into a toilet, and promptly flushing and pouring bleach everywhere. FYI, Shithead is basically Clayface with bloodlust permanently on.
So, does The Killer stand a chance against all of Konoha, by himself?

Here's the thing: He's still a normal human in all physical aspects. He has no superstrength, superspeed, superendurance. He wears basic combat gear, nothing flashy besides his flight boots. His guns are just regular pistols, not SoK guns.

I'd say Konoha's only chance is to overwhelm him.

I own the Wanted limit series and I would equate the killer to being along the lines of the bullseye I've read. Hence, if you are indeed good enough, and you need to be pretty damn good, you can avoid him. Basically then, any ninja with at least daredevil overall abiility/movibilaity should be able to dodge a couple of shots... so basically any high end chunin at worst.
in terms of comparing The Killer to bullseye, The killer deflected a bullet from point blank range with a knife. The killer can hit targets at ridiculous angles and speed, through what I would assume we could explain as some kind of algorithmic subconcious ability. However, aside from speed, Wesley Gibson is only along the lines of perhaps peak human in everything else. In short, he doesn't have close to what it takes to stand a chance. Hell, even if we use the feat of his father running along side a building, he is still not beyond peak in anything but some types of speed. So, just because he has the flight boots doesn't mean he is suddenly unstoppable.
In fact, what we have here is a Deidara like character attacking Konaho. Accept in this case, the character has slower reaction speed/ combat speed, less durability/strength, less destructive power, is slower in the air as fair as I'm concerned, perhaps less maneverability in the air (although there isn't much to compare), and fair better accuracy... but with a more specific target zone... of which hurts him given the speed of anyone decent in the village. Further still, The Killer has no knowledge of any of the various jutsu's that will come at him, and equally so, he will be hard pressed to adapt to them and or survive. On top of all this, the Hyuua, I.E> Hiashi = the perfect counter. Another thing is the fact that other individual Jounin could likely rape him. Not to mention he has no counter or idea about genjutsu. finally then, I presume the only positive argument for the Killer, and the making of this thread is on the basis of the ninja's being unable to dodge his gun fire. On this point, I think that's ridiculous and that any jounin has no trouble whatsoever... or simply does something in a jutsu far better then simple highpowered gun fire.
In conclusion The Killer loses horribly, and stands no chance whatsoever.

...There's an enemy called Shithead? Why have I not been notified of this?

The difference between Bullseye and The Killer is that while both have superhuman accuracy, Bullseye seems to prefer non-lethal objects for weapons (which he then turns lethal) as opposed to The Killer, who sticks with guns. I doubt a Naruto ninja could dodge modern pistol shots from a guy who can take out the wings of a fly while not even aiming.
