if you have photoshop all you have to do is duplicate the first layer- delete the background [not the background copy] and then use the magic wand. :D

You can only use this method if the background has a solid colour.
Save it as a png ,gifs make the file size smaller but lose quality.

If you're talking about tranparencies, when you can make a new layer, put the shape or whatnot in the sig, and then create a clipping mask.
